मंधाना का जन्म 18 जुलाई 1996 को मुंबई , महाराष्ट्र में स्मिता और श्रीनिवास मंधाना के घर एक मारवाड़ी हिंदू परिवार में हुआ था। उनके पिता एक रासायनिक वितरक के रूप में काम करते थे, जबकि उनकी माँ एक गृहिणी थीं।  जब वह दो साल की थी, तो उसका परिवार महाराष्ट्र के सांगली के एक उपनगर माधवनगर में चला गया , जहाँ उसने अपनी स्कूली शिक्षा पूरी की। उन्होंने सांगली में चिंतामन राव कॉलेज ऑफ कॉमर्स में पढ़ाई की। मंधाना के पिता सांगली के लिए जिला स्तर पर क्रिकेट खेलते थे, जैसा कि उनके भाई श्रवण ने किया था, जो अब एक बैंक मैनेजर हैं। अपने भाई को महाराष्ट्र राज्य अंडर -16 टूर्नामेंट में प्रतिस्पर्धा करते देख मंधाना को इस खेल को अपनाने की प्रेरणा मिली। नौ साल की उम्र में, वह महाराष्ट्र की अंडर -15 टीम के लिए चुनी गई थी |

When she was two years old, her family moved to Madhavnagar, a suburb of Sangli, Maharashtra, where she completed her schooling. She studied at Chintaman Rao College of Commerce in Sangli.
